چکیده

Osteosarcoma (OS), a severe malignant bone tumour, usually occurs in adolescents and children has poor prognosis. Asiatic acid (AA), an active component isolated from Centella asiatica (L.) Urb., exhibits appreciable anti-oxidant anti-tumour activities. So far, the effects underlying mechanisms of AA against OS have not been clarified. Here, we explored human involved mechanism mediating its actions. To evaluate on cell proliferation cells, viability colony formation assays were performed. Flow cytometry was used to apoptosis cells exposed mitochondrial membrane potential. Western blotting RT-PCR applied determine expression relevant proteins their mRNA levels. Our explorations showed that inhibits concentration- time-dependent manner, induces by intrinsic (mitochondrial) pathway. Importantly, found inhibition AA-induced phosphorylation JAK2/STAT3 signalling molecules decrease MCL-1 contributed efficacy AA. Collectively, our results suggest could evoke mitochondrial- induced suppression pathway expression. These strongly demonstrate be potential agent for treatment.

Millimeter wave treatment induces apoptosis via activation of the mitochondrial-dependent pathway in human osteosarcoma cells.

Millimeter wave (MW) is an electromagnetic wave with a wavelength between 1 and 10 mm and a frequency of 30-300 GHz that causes multiple biological effects and has been used as a major component in physiotherapies for the clinical treatment of various types of diseases including cancers. However, the precise molecular mechanism of the anticancer a...

Inhibition of JAK2/STAT3 signalling induces colorectal cancer cell apoptosis via mitochondrial pathway

Abnormalities in the JAK2/STAT3 pathway are involved in the pathogenesis of colorectal cancer (CRC), including apoptosis. However, the exact mechanism by which dysregulated JAK2/STAT3 signalling contributes to the apoptosis has not been clarified. To investigate the role of both JAK2 and STAT3 in the mechanism underlying CRC apoptosis, we inhibited JAK2 with AG490 and depleted STAT3 with a smal...

Bufalin induces apoptosis in the U‑2OS human osteosarcoma cell line via triggering the mitochondrial pathway.

Bufalin has been shown to induce apoptosis in osteosarcoma cells; however, the underlying mechanism has not been elucidated. The purpose of the present study was to investigate whether mitochondria‑mediated signaling pathways trigger the process of apoptosis in the U‑2OS osteosarcoma cell line. Bufalin inhibited the proliferation and induced apoptosis in U‑2OS cells in a time- and dose‑dependen...

Celastrol induces apoptosis of human osteosarcoma cells via the mitochondrial apoptotic pathway

Celastrol is an active compound extracted from the root bark of Triptergium wilfordii Hook F., also known as ̔Thunder of God Vine̓. It is a well-known Chinese medicinal herb that was found to inhibit tumor cell growth and promote apoptosis in several tumor cell lines. However, research into its effects on osteosarcoma cell apoptosis is still extremely limited. The present study was undertaken to ...

AICAR induces mitochondrial apoptosis in human osteosarcoma cells through an AMPK-dependent pathway

The AMP-activated protein kinase (AMPK) activator 5-aminoimidazole-4-carboxamide ribonucleotide (AICAR) modulates cellular energy metabolism, and promotes mitochondrial proliferation and apoptosis. Previous studies have shown that AICAR has anticancer effects in various cancers, however the roles of AMPK and/or the effects of AICAR on osteosarcoma have not been reported. In the present study, w...
